The insert contains the following restriction sites (kb from the 5' end): BglI--3.03; BglII--3.1; EcoRI--4.33; PstI--2.2, 2.4, 3.23, 3.28; PvuII--3.21, 3.28, 4.17; SacI--2.43, 2.58, 3.0; SmaI--2.33. An 0.8 kb PstI fragment of the insert hybridizes to the following fragments of human genomic DNA (kb): EcoRI--4.7; HindIII--7.2; BamHI--10.4; BglII--4.0; StyI--1.3. Detects a 9.0 kb EcoRI fragment of hamster genomic DNA. Contains a complete exon 2 (2.5 kb) plus upstream and downstream sequences. Exon 2 contains most of the 5' untranslated region, the complete coding sequence (nt 2182-3519 of the sequence record) and the complete 3' untranslated region. Derived from pHG15 which was constructed by inserting a SalI fragment from an lambdaEMBL3 library into pGEM-5Z. pHG4.5 is a circularized NcoI fragment from pHG15, and thus retains lambdaEMBL3 sequences from SalI to the 5' end of the insert. The SP6 promoter is adjacent to the 5' end of the insert. |
